Primer walking is a method of for sequencing DNA fragments between 1.3 and 7 kb. . The DNA of interest may be either a plasmid insert or a PCR product. The initial sequencing will be performed from each end using either universal primers or designed primers. In order to completely sequence the region of interest Sequencher will design and synthesize primers as necessary to obtain coherent sequence information.We offer following services.
SSPW – Single Stranded Primer Walk
Single strand primer walks provide contiguous sequence information for one strand of the region of interest. Since only one strand is sequenced we can only guarantee an accuracy level of between 95 and 98 percent. The customer will receive the sequence as well as the contig map of all sequencing performed to cover the region.
DSPW – Double Stranded Primer Walk
Double strand primer walks provide contiguous sequence information for both strands of the region of interest. By sequencing both strands, the sequence will have an accuracy of 99.99 percent. The customer will receive the sequence as well as the contig map of all sequencing performed to cover the region.
