DNA sequencing encompasses biochemical methods for determining the order of the nucleotide bases, in a DNA oligonucleotide.DNA sequence determines the patterns that make up genetic traits and in some cases behaviors. Samples may be submitted in form of cells (glycerol stocks, stabs, plates) or DNA (plasmids, PCR products, BAC/PAC, lambda, cosmids/phosmids).Customers may either provide sequencing primers or provide an e-copy of primer sequences to be synthesized using Sequencher’s Ologonewciotide Sequencing Service. We provide 32 universal primers common to most commercial vectors for free of cost.
Sequencher also provide services for difficult template type such as:
- GC-rich
- 2° structures
- AT-rich
- PolyA, PolyG
- Repetitive regions
- Heterozygote insertions & deletions
Samples should be submitted according to the below guideline:
SampleType | Sample Concentration | SampleVolume | Primer Concentration | PrimerVolume |
---|---|---|---|---|
PCR | 100 ng/µl | 10 µl/reaction | 10 µM | 10 µl/reaction |
Genomic DNA | 300 ng/µl | 10 µl/reaction | 10 µM | 10 µl/reaction |
Plasmid | 300ng/µl | 10 µl/reaction | 10 µM | 10 µl/reaction |
Cosmid, BAC | 500 ng/µl | 10 µl/reaction | 10 µM | 10 µl/reaction |
